Since the first reported outbreak of dengue hemorrhagic fever in Pakistan, several mini outbreaks have been observed in the country. In Pakistan, Serotype 2 (DEN-2) is documented since 1994 and is the major circulating serotype in the region. Dengue virus serotype 3 (DEN-3) was first reported in Karachi during 2005 outbreak. Dengue virus has now become a serious threat to humans especially in developing countries. As yet, no vaccine and proper treatment is available for dengue virus, so control of vector is the only option left to control dengue infection. Aedes mosquitoes are well known vector for this virus but some studies show that dengue virus can also survive in Culex mosquitoes. So, we have conducted a study to compare the vectorial role of Aedes and Culex in Lahore by using molecular techniques such as PCR and nucleotide sequencing of the C-prM gene junction of Dengue virus.A total of 1,110 adult resting mosquitoes were collected from selected localities of Urban Lahore. Of collected Aedes (n=498) and Culex (n=612) each were divided into 45 pools separately. After molecular analysis, 6 pools of Aedes (13.33%) and 9 pools of Culex (20%) were found positive for dengue virus. Interestingly, in all positive pools only serotype-2 of dengue virus was detected.We have found only serotype-2 of dengue virus in our study but other studies also report the presence of serotype-3. The occurrence of serotype-1 and serotype-4 in Pakistan, till now, is not reported. The presence of dengue virus in Culex in such a high per cent is alarming.
